مقایسه روش سنجش آنزیمی با روش تخمیر چند لوله‌ای در ارزیابی کیفیت میکربی آب رودخانه کارون

چکیده

به‌منظور طراحی صحیح و ارزیابی کارایی واحدهای تصفیه آب لازم است پایش میکربی آبهای سطحی که به‌عنوان منابع آب آشامیدنی تلقی می‌شوند، مورد توجه قرار گیرد. سنجش آنزیمی رویکردی است که به‌منظور ارزیابی سریع کیفیت میکربی منابع آبی به‌کار می‌رود. در این مطالعه برای بررسی کیفیت میکربی آب رودخانه کارون، روش سنجش آنزیمی با استفاده از محیط مایع LMX با روش متداول تخمیر چند لوله‌ای MTF مقایسه گردید. میانگین شمارش شده کلیفرم‌های کل به‌روش LMX و MTF به‌ترتیب برابر با MPN/100ml 9928 و 7564 و برای اشریشیاکلی MPN/100ml 6684 و 6546 بود. آنالیز آماری، این اختلاف را برای برآورد کلیفرم‌های کل، معنی‌دار نشان داد اما در مورد اشریشیاکلی اختلاف معنی‌داری حاصل نگردید. با توجه به یافته‌های این مطالعه به‌نظر می‌رسد که LMX روش مناسبی برای ارزیابی و شمارش کلیفرم‌های کل و اشریشیاکلی در آبهای سطحی است. سرعت، سادگی و شمارش همزمان کلیفرم‌های کل و اشریشیاکلی از مزایای این روش محسوب می‌گردد.

چکیده [English]

Microbiological monitoring of surface waters designated for use as drinking water is essential by water utilities for the design and operation of drinking water treatment plants. Enzymatic assays have been applied as a rapid alternative approach to assess the microbiological quality of freshwater. In this study, the LMX broth (LMX) as an enzymatic assay was compared with the standard method of multiple tube fermentation technique (MTF) for the microbial monitoring of the Karoon River. Enumeration of total coliforms and E. coli averaged 9928 and 6684 MPN/ 100 ml by the LMX and 7564 and 6546 MPN/ 100 ml for the MTF, respectively. This difference was statistically significant for TC but the overall analysis revealed no difference between E. coli recoveries on LMX and MTF. In conclusion, LMX can be used for the enumeration of coliforms and E. coli in surface waters as it is less lobar-intensive, yields faster result, and simultaneously detects both total coliforms and E. coli.
