جداسازی، شناسایی و تعیین خصوصیات یک گونه مخمر مولد آمیلاز و کاربرد آن در تولید کاروتنوییدها با استفاده از کشت توأم

چکیده

نشاسته پلی ساکاریدی گیاهی است که طی سالهای اخیر به دلیل افزایش تولید و فرآوری آن در ایران، پساب حاصل از آن از آلاینده‌های زیست محیطی محسوب می‌گردد. در این تحقیق، پس از جداسازی یک گونة مخمری مصرف کنندة نشاسته (مولد آمیلاز) و شناسایی آن به عنوان کریپتوکوکوس آیروس، برخی خصوصیات آن از لحاظ الگوی ترشح آمیلازها ، نحوه تجزیة نشاسته، ویژگی آمیلازهای مترشحه و قابلیت آن در تولید کاروتنوییدها با استفاده از کشت توأم مورد بررسی قرار گرفت. طبق نتایج به دست آمده مخمر کریپتوکوکوس آیروس قادر به تجزیه و مصرف نشاستة محلول و نشاستة خام ذرت است؛ مصرف نشاستة خام در میان مخمرها جزء صفات نادر بوده و از لحاظ صنعتی اهمیت ویژه‌ای دارد. همچنین مخمر مولد آمیلاز قادر به تجزیه کامل نشاستة محلول در طی مدت 10 ساعت اولیه کشت و تبدیل کامل آن به مجموعه‌ای از قندهای احیا کننده است و لذا با توجه به دسته‌بندی مخمرها از لحاظ الگوی ترشح آمیلازها، این مخمر جزء مخمرهای تند رشد بر روی نشاسته محسوب می‌شود. عدم بیماری‌زایی، رشد مناسب این گونه بر روی نشاستة خام و محلول، مقاومت دمایی مناسب (کمتر از °C55) مجموعة آمیلازهای مترشحه و نیز pH مطلوب حدود 6-5/5 برای فعالیت مجموعة آمیلازهای مترشحه توسط مخمر کریپتوکوکوس آیروس، بهره‌گیری از این مخمر را در فرآیندهای صنعتی برای تولید محصولات میکروبی میسر می‌سازد. در کشت توأم مخمر کریپتوکوکوس آیروس به همراه یک گونه از مخمر رودوتورولا به عنوان مخمر مولد کاروتنوییدها که به عنوان مرحله آزمایشگاهی تولید کاروتنوییدها انجام گرفت، طیف جذبی کاروتنوییدهای تولید شده در w/v%3 نشاستة محلول  بررسی گردید و مشخص شد که کیفیت کاروتنوییدهای تولید شده نسبت به کشت خالص مخمر رودوتورولا، تغییری نیافته است.

چکیده [English]

Starch is a plant polysaccharide with unique applications in Iran. Its increasing production and processing recently have led to large volumes of industrial effluent as an environmental pollutant. In this study, an amylase producing yeast is isolated and identified as “Cryptococcus aerius” to investigate some of its characteristics such as its amylase secretion and starch digesting patterns, kinetics of amylase complex, and its capability for carotenoid production in dual culture. The results indicate that C.aerius is capable of soluble and raw maize starch digestion and assimilation. Raw starch digestion is scarce among yeast species; hence, it is industrially important. C.aerius digests soluble starch in the first 10 hours of cultivation and on the basis of amylase secreting patterns, it is therefore categorized with fast growing species on starch as carbon source. Non-pathogenicity, digestion of raw starch, heat stability of the secreted amylases complex (>55˚C), and the optimum pH level of 5.5- 6 for amylases complex are the set of properties that make this species capable of use in microbial production on an industrial scale. Absorption of carotenoid extract obtained from dual fermentation of C.aerius and Rhodotorula sp. indicates that the quality of carotenoids produced in dual fermentation is the same as that produced from pure Rhodotorula sp culture.
